1. Two color immunohistochemistry for Ki 67 nuclear brown with Red LCA with Hematoxylin counterstain (20X).
2. Auto-counting and auto-marking as first step with KoShiPath AI software. RED dots mark Ki-67 immunoreactive nuclei as POSITIVE with GREEN dots mark Ki-67 npn-immunoreactive nuclei as NEGATIVE.

3. Manual adjustment is possible with KoShiPath for correcting some false-positively and false-negatively marked / counted tumor nuclei, based on interpreter skill with additional input based on two color immunohistochemistry (Ki 67 nuclear brown, LCA cytoplasmic Red) allowing to confirm lymphocyte/leucocyte nature of positive (Red arrows) and negative (Green arrows) cells and weed them out of the calculation by unmarking those cells manually.

If these cells are not properly excluded the Ki-67 index generated would be inaccurate compromising the grading (Grade 1: 0 to 3%; Grade II: 4 to 20%; Grade III: >20%) of tumor, especially those in borderline category between Grade I / Grade II (Ki-67 in the range of 0 to 6 %) and Grade II / Grade III (Ki-67 in the range of 15 to 25 %).
